CureVac fails in pivotal COVID-19 vaccine trial with 47% efficacy
    Ludwig Burger

June 16 (Reuters) - German biotech CureVac NV (5CV.DE) said on Wednesday its COVID-19 vaccine was only 47% effective in a late-stage trial, missing the study’s main goal and throwing in doubt the potential delivery of hundreds of millions of doses to the European Union.

The disappointing efficacy of the shot known as CVnCoV emerged from an interim analysis based on 134 COVID-19 cases in the study with about 40,000 volunteers in Europe and Latin America.

The stakes for CureVac and prospective buyers of its vaccine in Europe had risen after age limits were imposed on the use of the Johnson & Johnson (JNJ.N) and AstraZeneca (AZN.L) vaccines due to a link to extremely rare but potentially fatal clotting disorders.

CureVac's shot was also expected to help in low and middle-income countries that have lagged far behind richer nations in the global immunisation drive.

As CureVac’s only major supply deals, the European Union in November secured up to 405 million doses of the vaccine, of which 180 million are optional. That was followed by a memorandum of understanding with Germany for another 20 million doses.

CureVac's U.S. traded shares fell 50.6% to $46.81 in after-hours trading following publication of the data.

The company said at least 13 virus variants accounted for the infections among the study population.

Out of the reported COVID-19 cases in the trial, 124 were sequenced to identify the variant causing the infection, it said. One case was attributable to the original version of the SARS-CoV-2 coronavirus that emerged in Chinese city of Wuhan in late 2019, while 57% of the cases were caused by more highly transmissible so-called variants of concern.

The company added that the interim results suggest the vaccine is effective in younger participants but did not prove efficacy in those above age 60, the age group most at risk for severe COVID-19.

"While we were hoping for a stronger interim outcome, we recognize that demonstrating high efficacy in this unprecedented broad diversity of variants is challenging. As we are continuing toward the final analysis with a minimum of 80 additional cases, the overall vaccine efficacy may change," said Chief Executive Franz-Werner Haas.

Dr. Amesh Adalja, in infectious disease expert at the Johns Hopkins Center for Health Security, said the variants did not completely explain the efficacy number and he still wants to see data specifically about CureVac's ability to stop serious disease, hospitalization and death.

"If it can do that, even with a 47% efficacy in symptomatic disease, that's still a very valuable thing. That's all we've ever wanted vaccines to do," he said.

Dr. Peter Hotez, a virologist and dean of the National School of Tropical Medicine at Baylor College of Medicine, said it was not clear if the issue was variant-specific or the vaccine's inability to create high levels of neutralizing antibodies.

Since starting out in 2000, Tuebingen-based CureVac has focused on so-called messenger RNA (mRNA) technology, which is also behind the success of BioNTech (22UAy.DE) and partner Pfizer (PFE.N) as well as Moderna (MRNA.O), whose vaccines have proved more than 90% effective in preventing illness.

Prior to the emergence of the highly effective vaccines, the U.S. Food and Drug Administration had targeted at least 50% efficacy, which CureVac has fallen short of in the interim analysis. The World Health Organization had said it was looking for at least 70% efficacy.

While late-stage trials of the BioNTech/Pfizer and Moderna vaccines were conducted when the original version of the virus was dominant, real-world data has so far suggested only somewhat weaker protection against the new variants.

CureVac, which is backed by investors Dietmar Hopp, the Gates Foundation and GlaxoSmithKline (GSK.L), as well as the German government, had aimed to produce up to 300 million doses of the vaccine in 2021 and up to 1 billion doses in 2022.

Manufacturing partners include Celonic Group of Switzerland, Novartis (NOVN.S), Bayer (BAYGn.DE), Fareva, Wacker (WCHG.DE) and Rentschler Biopharma SE.

GSK has agreed to collaborate with CureVac on production as well as on development of the next-generation vaccines. A company spokesman said early, preclinical data indicated a 10-fold increase in immunogenicity versus the first.

"We aim for availability of a second-generation COVID-19 vaccine as early as the second half of 2022, subject to regulatory approvals," he said.

Matt Hancock quits as health secretary after breaking social distance guidance

    By Emma Harrison

Matt Hancock resigned as health secretary following pictures being published of him in an embrace with Gina Coladangelo (left)

Matt Hancock has resigned as health secretary after he breached social distancing guidance by kissing a colleague.

In a letter to the PM he said the government "owe it to people who have sacrificed so much in this pandemic to be honest when we have let them down".

Boris Johnson said he was "sorry" to receive the resignation.

It comes after pictures emerged of him with Gina Coladangelo, reportedly taken on 6 May.

Mr Hancock had been under increasing pressure to quit, after The Sun published pictures of Mr Hancock and Ms Coladangelo, who are both married with three children, kissing. The newspaper said they had been taken inside the Department of Health.

Fellow Tory MPs, as well as Labour and the Covid-19 Bereaved Families for Justice group, had called for the health secretary to be sacked.

BBC political editor Laura Kuenssberg said No10 had stressed that it had been Mr Hancock's decision to go and that he had not been pushed out by the prime minister.

She said Ms Coladangelo had also left her role as a non-executive director of the Department of Health.

In a video posted on Twitter, Mr Hancock said: "I have been to see the prime minister to resign as secretary of state for health and social care."I understand the enormous sacrifices that everybody in this country has made, that you have made, and those of us who make these rules have got to stick by them and that's why I have got to resign."

    20 năm thất bại.

Cái giá phải trả: 59 lính tráng tử vong, 12 tỉ thuế của dân Đức.

    • By the end of 2020, the Bundeswehr's deployment had cost the German taxpayers around €12.5 billion ($15 billion).
    • The mission, which lasted for almost 20 years, cost the lives of 59 German soldiers

Germany pulls last troops from Afghanistan, ending nearly 20-year mission
Germany has finished withdrawing its last contingent of around 570 soldiers from Afghanistan as the security situation deteriorates. The move marks the end of a nearly 20-year mission.

After nearly 20 years, Germany's Afghanistan mission has officially ended

With US troops eyeing their final exit from Afghanistan in September, Germany pulled out all of its remaining troops on Tuesday. Last week, Germany's Defense Ministry said around 570 troops were still deployed.

The last of them have now been flown out of the northern city of Mazar — ending a nearly two decade-long mission. The contingent also included members of the KSK special forces, who were tasked with securing the camp during the move.

German Defense Minister Annegret Kramp-Karrenbauer said the soldiers were "now on their way home."

"This marks the end of a historic chapter – an intense deployment that has challenged and shaped us," she wrote on Twitter.

She thanked the soldiers for fulfilling their duties in Afghanistan "with professionalism and conviction."

Looking back at the mission, the minister pledged to discuss "what was good, what was not good and what we've learned."

Germany's armed forces also confirmed the pullout, saying that "last soldiers have left Afghanistan."

The German military had kept the details on the pullout vague on security grounds. Speaking just hours before the move was officially confirmed, Kramp-Karrenbauer said the withdrawal was happening "in an orderly manner, but also as swiftly as possible." Germany maintained a contingent of around 1,100 troops before starting the drawdown in May.

    What happens next?

The four military planes carrying the troops are expected to make a stop in Tbilisi before landing in Germany on Wednesday. Their old camp will be handed to Afghani troops as Germany ends its participation in NATO's "Resolute Support" mission.

After landing in Germany, the soldiers will enter a mandatory two-week quarantine to curb the risk of the highly contagious Delta coronavirus variant. According to Der Spiegel magazine, at least one German soldier was infected with the variant earlier this month.

How long were German troops in Afghanistan?

Germany deployed its forces in the wake of the deadly 9/11 attacks on the US conducted by al-Qaida in 2001. The first troops arrived in Kabul in January 2002.

In the early stages of the mission, German soldiers were told that their deployment was aimed at stabilizing the country rather than fighting the Taliban.

Over 150,000 German soldiers have been stationed in Afghanistan over the years, many of them serving more than once.

By the end of 2020, the Bundeswehr's deployment had cost the German taxpayers around €12.5 billion ($15 billion).

The mission, which lasted for almost 20 years, cost the lives of 59 German soldiers.

It was the longest, most expensive and the bloodiest German deployment since WWII. According to UN data, the Afghanistan conflict claimed the lives of over 39,000 civilians since 2009. While most of them have been killed by the Taliban, international forces also caused civilian causalities, especially through air strikes. The US also lost 2,442 soldiers since the mission started.

    What is happening in Afghanistan?

The end of the mission comes as the Taliban in Afghanistan rapidly seize new territory from the US-backed government in Kabul. Taliban forces are also positioning themselves around provincial capitals, prompting fears that they will seize full control once all foreign troops leave. The UN reported a spike in civilian casualties since last September.

The extremist Islamic militants were notably emboldened by the US pledge to finish its Afghanistan withdrawal this year. The administration of Donald Trump had pledged to complete the pullout by May this year. His successor, Joe Biden, changed the target date to September 11 this year, the 20th anniversary of the attacks which prompted the conflict.

While meeting with Afghanistan President Ashraf Ghani on Friday, Biden promised a "sustained" partnership between the countries. However, he did not signal any changes to the timetable due to the Taliban offensive.

"Afghans are going to have to decide their future," Biden told reporters.
